ABSTRACT
Abstract: This study explores optimizing drug delivery by integrating microneedle (MN) technology with iontophoresis (ITP), focusing on transitioning from traditional cone-shaped MNs. The aim is to assess how modified MN geometries, particularly MNs with barbed structures, influence electric field distribution and surface area, impacting drug delivery efficiency. Projected outcomes anticipate increased model drug penetration using modified MNs with ITP, signaling potential for superior delivery across gel layers. Comparative analyses reveal penetration depth differences between modified and cone-shaped MNs, providing critical insights into delivery efficacy.
Simulations via COMSOL Multiphysics demonstrate that modified MNs exhibit enhanced electric field strengths, especially at their edges, augmenting drug concentrations within the skin. Results indicate that modified MNs achieve higher electric field strengths than cone-shaped MNs, primarily concentrated at their edges. Moreover, modified MNs create longer channels compared to traditional MNs, suggesting greater potential for drug penetration. Experimentation using drug model and agarose gel as a skin model corroborates these findings, showcasing improved drug penetration and diffusion through microholes created by modified MNs, potentially advancing delivery techniques. This investigation encompasses design, fabrication, experimental analysis, and discussions on MN and ITP interactions, aiming to refine delivery through optimized MN structures and ITP integration.
